An odyssey of profanity, a bombardment of foul language, culminating with a mock-Disney tune in the form of Uncle Fucker. South Park: Bigger, Longer & Uncut is similarly shocking, but that scene is a sucker punch you're never quite prepared for.
I like South Park as much as the next guy and Uncle Fucker is one of the catchiest ditties ever penned for a feature film, but what's so big-screen essential about this? South Park works equally well on the small screen, from whence it came.

Seeing that on the big screen, and being as shocked as everyone else in the audience was definitely big screen stuff.

If you say it Punch, then it must be true. I never had the pleasure of seeing it on the big screen only on DVD. I guess I'm most used to seeing SP on my telly.

"I I believe in America." - the opening scene of The Godfather. It's a haunting opening monologue. Then Brando asks:
"Why did you go to the police? Why didn't you come to me first? " - and the whole setup is complete and defined.

The shot of Andy standing tall and proud following his escape where he has just trudged through the sewage and the rain has wiped it all off. His s**t dont stink!

Smile Inglourious Basterds-

The ending where Shosanna's message is played on the screen as the theater burns. It was so much better at the cinema than it looks on a TV.

The scene in Casino Royal where a certain Mr Craig is tied to a chair and is shall we say 'whipped', every man in the screen crossed his legs and winced.

Default For sheer bloody excitement

The Hit Girl slaughter scenes in Kick-Ass. Much better on the big screen than on DVD.

Default 2001: A Space Odyssey

When scientists on moon discover the Monolith, which is almost same ratio as a cinema screen. A high pitched signal is triggered when Floyd touches it: in a sense Kubrick triggers a leap in understanding in the audience, through the scientists in the film screen, breaking the fourth wall.

Nice thread.

I'm goona mention a few that already have been, but what the hey!!

Bullet Time in The Matrix, just blew me away, completely left the cinema it blew me away so much. (ooo-er)

Many moments in the LOTR, The Balrog, Helms Deep and definatly Pellanor Fields.

King Kong vs V-Rex smack down, the film may of been a tad, um too long, but that 20 mins was fapping brilliant.

Many moments in Transformers, mention goes to when Barricade barrels forward, transforms in mid-air, then boots it after bumblebee, geeky, but fucking awesome.

Indiana Jones and the Crystal Skull, hang in there with me...I was too young to cath the first two, and never got to see the third at the cinema for reasons I cant remember, but to finally hear that score coming at me from the big screen was worth the ticket...shame about the following too far fetched, alien crap, gopher infested 2 hours that followed.

Jurrasic Park, oft forgotten "summer film". It may of been on TV a trillion times, but I still remember the first time I saw that T-Rex in full glory, brilliant stuff.

Avatar. Oh, 3D can work.

The Dark Knight, plenty of moments, Bank Robbery, Truck Flip, Pencil Trick, any moment Heath Ledger is on screen.

Lots more...
